What do we actually know about NMN supplementation and its effects on NAD+ levels, sirtuins, and biological age?
Why NAD+?
NAD+ (nicotinamide adenine dinucleotide) is a central coenzyme in essentially every cell, essential for mitochondrial energy production, DNA repair, and sirtuin activity — a family of enzymes regulating gene expression, stress responses, and metabolism.
NAD+ levels decline measurably with age — in some tissues by up to 50% between young adulthood and age 70.

NMN as a precursor
NMN (nicotinamide mononucleotide) is a direct precursor to NAD+. The hypothesis: supplementing NMN raises NAD+ levels and activates sirtuins.
What the evidence shows
- Animal models: Robust improvements in mice — better mitochondrial function, insulin sensitivity, endurance.
- Human studies (Phase I/II): Several show that oral NMN does raise blood NAD+ levels. Functional endpoints (walking speed, sleep quality, aerobic capacity) improve modestly in some.
- Limitations: Studies are small (often under 100 participants), short (4–12 weeks), and frequently industry-funded. No long-term outcome data exists yet.

My take
NMN is one of the most mechanistically plausible longevity supplements. But: it's expensive, clinical evidence is early-phase, and the fundamentals (sleep, strength training, protein, zone 2) offer a much bigger lever for most people.
If you've covered the basics and want to experiment: defensible. If you're not yet exercising regularly: the money is better spent on a gym membership.
